Zusammenfassung
In this study investigates peroxidases from Baltic Sea macroalgae with regard to their identification, recombinant production and biotechnological potential. In contrast to well-studied plant peroxidases, algal peroxidases remain largly unexplored, particularly with respect to their function. Transcriptomic data, native protein purification, and mass spectrometry were combined to identify peroxidases from the macroalgae. Three candidates were selected for heterologous expression in E. coli. Two candidates were successfully produced and biochemically characterized following cofactor reconstitution. A cytochrome c peroxidase from the green alga Bryopsis plumosa was analyzed with respect to activity and stability, while a vanadium-dependent haloperoxidase from the red alga Ahnfeltia plicata, including a truncated variant, was applied in a luminol-based chemiluminescence assay.
