Item-typ:Veröffentlichung, Untersuchungen zur Struktur und Regulation des Photosyntheseapparates in Cyanobakterien(2010-04-13); ; ; Gloeobacter violaceus (Gloeobacter) is a slow-growing unicellular cyanobacterium. A unique feature of Gloeobacter is the lack of thylakoid membranes. Gloeobacter has been refractory in terms of genetic manipulation. Chapter 1 describes a transformation system for Gloeobacter facilitating its genetic manipulation. The successful transformation system was then utilized in chapter 2. The aim was to study the effect of heterologous vipp1 expression on the membrane structure of Gloeobacter, which lacks Vipp1. Vipp1 appears to be essential in thylakoid formation and shares sequence homology with the bacterial phage shock protein A (PspA). Expression of vipp1 from Anabaena sp. PCC 7120 or an artificial vipphybrid gene led to improved vitality of Gloeobacter. Electron micrographs revealed that the presence of the Vipphybrid or the Vipp1 protein is not sufficient to induce changes in membrane structure. Chapter 3 deals with regulatory aspects of the photosynthetic apparatus of cyanobacteria. In plants and green algae PSII core proteins undergo light dependent phosphorylation. This reversible phosphorylation is absent in cyanobacteria, although the phosphorylation sites are conserved. The stn8 gene of Arabidopsis thaliana, encoding the kinase specifically phosphorylating PSII, was expressed in Synechococcus PCC 7942. STN8 catalyzes the phoshorylation of the D1 protein in Synechococcus as revealed by immunodecoration. The phosphorylation of the D1 protein in Synechococcus led to increased growth rates, higher chlorophyll contents and differences in fluorescence emission spectra at 77K when cells were grown under suboptimal temperature.Dissertation262 168 - Some of the metrics are blocked by yourconsent settings
Item-typ:Veröffentlichung, Investigation of the role of class I KNOX genes in the floral development of Eschscholzia californica(2010-08-09); ; ; Four class I KNOX genes were isolated from the Eschscholzia californica, Papaveraceae. Phylogenetic analysis suggests that these four KNOX genes cover all three for the model Arabidopsis thaliana established clades of class I KNOX genes, STM (SHOOT APICAL MERISTEM-LESS), KNAT1/BP (Knotted-like Arabidopsis thaliana-1/BREVIPEDICELLUS) and KNAT2/6 (Knotted-like Arabidopsis thaliana-2/6). Two of these genes (EcSTM1 and EcSTM2) were identified as STM-like genes. Expression analysis in vegetative and floral shoots tips revealed a continuous expression of EcSTM1, EcKNAT1 and EcKNAT2, whereas EcSTM2 is exclusively expressed in early floral buds. EcSTM1-VIGS (Virus-induced gene silencing) resulted in the absence of the inner floral whorls. In flowers of EcSTM2-VIGS treated plants, the carpels were initiated but did not develop properly. Rarely, a floral reversion was observed. (Brunnenlebermoos):Identifizierung und Charakterisierung der sie codierenden Genfamilie(2002-02-08); ; ; Untersuchungen der Chlorophyll a/b bindenden (CAB) Lichtsammelproteine erstreckten sich bislang vorwiegend auf Samenpflanzen und einige wenige Algengruppen. Im Rahmen dieser Arbeit wurde die Zusammensetzung der cab-Genfamilie eines Bryophyten erstmalig detailliert untersucht.12 Mitglieder der cab-Genfamilie von Marchantia polymorpha wurden identifiziert. Neun der ermittelten cDNA-Sequenzen lassen sich Proteinen des Antennensystems des PSII zuordnen, eine Sequenz dem des PSI. Weiterhin konnten eine Teilsequenz für ein PsbS-Protein und ein Sep1-ähnliches Protein ermittelt werden.Die Zusammensetzung der cab-Genfamilie weist darauf hin, daß sich der Aufbau des LHCII aus den verschiedenen Lhcb-Proteinen seit mehr als 480 Millionen Jahren, seit den primitivsten Landpflanzen, nicht mehr gravierend verändert hat. Dafür spricht abgesehen von der Zusammensetzung der Genfamilie auch der hohe Grad an Sequenzhomologie zwischen den CAB-Proteinen von M. polymorpha und denen der Samenpflanzen.Im Vergleich zur Grünalge C. reinhardtii verfügt M. polymorpha über ein Lhcb2 und ein Lhcb3. Ein eindeutig dem Lhcb1 Protein Höherer Pflanzen zuzuordnendes Protein konnte hingegen nicht gefunden werden. Stattdessen wurden LHCII-Proteine gefunden (LHCII1.1-1.4), die eine Mischform aus Lhcb1- und Lhcb2-Protein Höherer Pflanzen darzustellen scheinen. Es liegt die Vermutung nahe, daß sich das Lhcb1 erst später aus einem solchen entwickelte. Sep-Proteine sind der Proteinfamilie der Elips zuzuordnen. Erstmalig konnte hier eine cDNA Sequenz für ein Sep1-ähnliches Protein in einer Niederen Pflanze nachgewiesen werden. Es scheint sich ebenfalls um ein Lichtstress-induziertes Protein zu handeln. Expressionsanalysen zeigen, daß einzelne cab-Gene von M polymorpha ebenfalls wie die der Angiospermen diurnal exprimiert werden. Transcription of the three psbA genes is differentially regulated by light. Under highlight conditions, the transcription of psbAI is repressed and that of psbAII and psbAIII is induced. The D1:1degradation products, mediated by the protease FtsH, may be involved in transcriptionregulation of psbAI by binding in the promotor region. Additionally, C-terminal breakdown products bind to asequence with high similarity to isiB, a gene which encodes flavodoxin, a substituent for ferredoxin underiron depleted conditions in cyanobacteria.Dissertation262 170
